چکیده

Purpose TO examine the ability of retinal pigment epithelial (RPE) cells to superantigen to T lymphoc e&t2 resent bacterial es. Methods RPE cells, er interferon gamma (INFg) treated or untreated, were coincubated with Jurkat T-cells, staphylococcal enterotoxin E (SEE), and PMA for 24 hours. Supernatants were collected for interleukin-2 (IL-2) assay. Raji B-calls were assayed for comparison. Binding of SEE to RPE cells was determined with biotinylated SEE which was detected with phycoerythin-conjugated streptavidin and bv flow cvtometry. Results On a per cell basis, INFG activated RPE cells induced an IL-2 release that was about l/20 of that amount stimulated by Raji cells. The antiaen presentina capacitv of RPE cells increased with-the-density of HLA-DR-epitops per cell. SEE bound only to HLA-DR positive, INFg treated RPE cells. Binding of SEE to RPE cells as well as the release of IL-2 was abolished bv antibodies to HLA-DR. HLA-DR negative RPE celis did not stimulate IL-2 release. Conclusions Only INFg activated RPE cells presented superantigens. Thus, RPE cells exposed to INFg have the capacity to bind and present bacterial superantigen to T cells and, both, may play a role in ocular inflammatory processes.

Tumor necrosis factor-alpha (TNF-α) is a pro-inflammatory cytokine produced by a variety of cells, including hematopoietic and non-hematopoietic cells, malignant cells, macrophages, B lymphocytes, T lymphocytes, natural killer cells, neutrophils, astrocytes, endothelial cells, and smooth muscle cells. TNF-α is a homo-trimeric molecular whose individual subunits are composed of antiparallel beta...

Effects of Secreted Mast Cell Mediators on Retinal Pigment Epithelial Cells: Focus on Mast Cell Tryptase

Numerous mast cells are present in the choroid, but the effects of mast cell mediators on retinal pigment epithelial (RPE) cells are not well understood. We investigated the influence of mast cell mediators on RPE cells in vitro, focusing on tryptase. Expression of receptors was examined by the reverse transcription polymerase chain reaction. We also assessed production of interleukin 8 and vas...

Immediate Exposure to TNF-α Activates Dendritic Cells Derived from Non-Purified Cord Blood Mononuclear Cells

Background: Tumor necrosis factor alpha (TNF-α) is a primary mediator of immune regulation and might be required in the early stages of DC development from CD34+ cells. However, details of optimal timing of exposure to TNF-α in DC development process in monocytes or non-purified hematopoitic cells are still lacking and clear benefits of this approach to the development of DCs remain to be valid...

اثر عصاره گیاه Dendrostellera lessertii بر ترشح TNF-α و تنظیم کاهشی گیرنده های آن بر سطح مونوسیتهای انسانی در محیط کشت

Background : We investigated the mechanism of action of Dendrostellera lessertii effects on decreasing size of adeno-carcinoma of rat intestine and breast tumors. This purpose followed by assessing its effects on TNF-α and TNF-α receptors on cultured human monocytes.  Material and method : The breast and intestine tumors were induced in rats by DMBA and DMH, respectively. Human monocytes isolat...

TNF-α Decreases VEGF Secretion in Highly Polarized RPE Cells but Increases It in Non-Polarized RPE Cells Related to Crosstalk between JNK and NF-κB Pathways

Asymmetrical secretion of vascular endothelial growth factor (VEGF) by retinal pigment epithelial (RPE) cells in situ is critical for maintaining the homeostasis of the retina and choroid. VEGF is also involved in the development and progression of age-related macular degeneration (AMD). We studied the effect of tumor necrosis factor-α (TNF-α) on the secretion of VEGF in polarized and non-polar...
